What happens when ring strain becomes a driving force for C–C bond activation?
1-Arylcyclobutanols are strained four-membered-ring compounds that offer valuable opportunities for C–C bond cleavage and ring-opening transformations. The inherent ring strain of cyclobutane provides a thermodynamic driving force for ring opening, while the hydroxyl group enables distinct activation pathways.
1-Arylcyclobutanols undergo ring-opening reactions through two main pathways:
(1) Transition-metal-catalyzed ring opening
(2) Radical-mediated ring opening
In transition metal-catalyzed reactions, deprotonation of the hydroxyl group generates a metal alkoxide intermediate, which undergoes β-carbon elimination to form a δ-oxoalkylmetal intermediate. Palladium (Pd) and rhodium (Rh) catalysts are commonly used in this type of transformation.
In radical-mediated reactions, alkoxy radicals undergo β-scission to release ring strain, producing δ-ketoalkyl radicals that participate in subsequent bond-forming reactions.
By accessing either organometallic or radical intermediates, 1-arylcyclobutanols provide complementary strategies for selective C–C bond cleavage and skeletal modification. Their distinctive reactivity makes them valuable building blocks for radical chemistry, transition-metal catalysis, synthetic methodology development, and complex molecule synthesis.
