Pyrazolotetrahydropyridine is a fused bicyclic heterocycle combining a pyrazole ring with a partially saturated six-membered nitrogen-containing ring. Its rigid, three-dimensional framework provides a useful structural motif for exploring molecular recognition and optimizing the properties of bioactive small molecules.
The pyrazolotetrahydropyridine scaffold is present in multiple emerging small-molecule therapeutics, such as elecoglipron (AZD5004/ECC5004)(https://www.thelancet.com/journals/lancet/article/PIIS0140-6736(26)00748-8/fulltext) and aleniglipron (GSBR-1290)(https://doi.org/10.1038/s41591-026-04476-6).
Aleniglipron is an oral small-molecule GLP-1 receptor agonist in clinical development for the treatment of obesity.
Elecoglipron is an oral, once-daily small-molecule GLP-1 receptor agonist being developed for weight management in adults with obesity or overweight, including those with type 2 diabetes.
