Breakthrough Biocatalytic Synthesis of Enlicitide (MK-0616) Marks a New Era for Oral PCSK9 Inhibitor Production

Enlicitide (MK-0616), Merck’s first-in-class oral macrocyclic peptide PCSK9 inhibitor, lowers LDL-C and treats atherosclerotic cardiovascular diseases. Having finished Phase III trials and obtained FDA priority review, it is set for a global launch in H2 2026.

Its complex triple-fused macrocyclic structure with 12 amide bonds and unnatural amino acids makes traditional chemical synthesis inefficient. The conventional 43-step process requires extensive protecting group manipulation, dilute cyclization and repeated purification, leading to high costs, large waste output and limited kilogram-scale production, restricting global supply.

Merck’s novel multi-enzyme cascade biocatalytic route, published in Science (May 7, DOI: 10.1126/science.aed8713), addresses these pain points. The synergistic engineered enzyme system completes fragment assembly, ligation and macrocyclization without protecting groups or repetitive purification, enabling stable kilogram-scale production with lower costs and waste, supporting Enlicitide’s commercialization and global supply.

As a professional chemical and biocatalysis service enterprise, ChemScene keeps close track of cutting-edge synthetic technologies for macrocyclic peptide drugs. Boasting mature custom synthesis, unnatural amino acid preparation and biocatalytic process development capabilities, ChemScene offers high-purity raw materials, intermediate customization and scalable process optimization services supporting the R&D and industrial production of Enlicitide and analogous PCSK9 inhibitor candidates.
